Description

Introduction to RPS7 Antibodies

RPS7 antibodies target the 40S ribosomal protein S7, a component of the small ribosomal subunit critical for protein synthesis. These antibodies are primarily used to study ribosomal biogenesis, developmental disorders, and diseases linked to ribosomal dysfunction .

Notes:

  • All antibodies detect endogenous RPS7 at ~22 kDa .

  • Optimal dilutions vary by application (e.g., 1:50–1:200 for IHC-P) .

4.1. Immunohistochemistry (IHC)

  • AKI Studies: Anti-RPS7 antibody (1:500 dilution) used in cisplatin-treated kidney tissues showed upregulated RPS7 expression correlating with ferroptosis .

  • Cellular Localization: RPS7 localizes to the cytoplasm and ribosomes, confirmed via subcellular fractionation .

4.2. Functional Studies

  • Ribosome Assembly: Mutant RPS7 proteins (Mtu and Zma) incorporate into ribosomes but disrupt rRNA processing .

  • Behavioral Phenotypes: Rps7 haploinsufficiency in mice causes working memory deficits, implicating RPS7 in CNS development .

Limitations and Future Directions

  • Species-Specific Differences: Murine Rps7 mutations do not replicate human DBA phenotypes, suggesting divergent roles across species .

  • Therapeutic Potential: Targeting RPS7 in AKI requires further validation of ferroptosis pathways .

Target Background

Function
Ribosomal Protein S7B (RPS7B) is a component of the ribosome, a complex cellular machinery responsible for protein synthesis. The ribosome consists of two subunits: the small ribosomal subunit (SSU) and the large ribosomal subunit (LSU). The SSU binds to messenger RNA (mRNA) and facilitates the translation of the encoded genetic information. The LSU contains the peptidyl transferase center (PTC), a catalytic site responsible for the formation of peptide bonds, thus polymerizing amino acids delivered by transfer RNA (tRNA) molecules into a polypeptide chain. Newly synthesized polypeptides exit the ribosome through a tunnel in the LSU, interacting with protein factors that regulate their processing, targeting, and membrane insertion. RPS7B plays a crucial role in the nucleolar processing of pre-18S ribosomal RNA and the assembly of ribosomes.
