Description

Research Applications of RSC30 Antibody

The antibody is widely used to investigate:

  • Chromatin remodeling mechanisms: Identifies RSC30 localization at DNA double-strand breaks (DSBs) via chromatin immunoprecipitation (ChIP) .

  • Protein interaction networks: Validates physical interactions with yKu80, Mre11, and Rsc1/2 in co-immunoprecipitation assays .

  • Gene expression profiling: Correlates RSC30 loss with downregulation of cell wall integrity genes (CRH1, FKS1) and spindle checkpoint pathways .

DNA Repair and Chromatin Dynamics

  • Non-homologous end joining (NHEJ):

    • rsc30Δ mutants exhibit a 2.4-fold reduction in CA insertions at DSB repair junctions and increased large deletions (>700 bp) .

    • Recruitment to DSBs depends on Mre11 and yKu70 .

  • Nucleosome remodeling:

    • RSC30-containing RSC complexes show reduced nucleosome sliding activity compared to RSC2 isoforms .

Functional Redundancy and Genetic Interactions

  • Suppression mechanisms: Overexpression of RSC3 rescues viability defects in rsc7Δ and rsc14Δ mutants .

  • Phenotypic traits:

    • rsc30Δ strains are viable but osmosensitive .

    • Synthetic lethality occurs when combined with rsc7Δ or htl1Δ mutations .

Expression and Regulation

  • Transcriptional impact:

    • RSC30 deletion downregulates 9% of yeast genes, including those involved in cell wall biogenesis (SLT2, CHS3) .

    • Upregulates stress-response pathways under DNA damage .

Future Directions

  • Therapeutic potential: Targeting RSC30-interaction domains could modulate chromatin dynamics in fungal infections.

  • High-resolution mapping: Advanced Cryo-EM studies are needed to resolve RSC30’s role in RSC-nucleosome interactions .

Target Background

Function
RSC30 Antibody is a component of the chromatin structure-remodeling complex (RSC), which plays a crucial role in transcription regulation and nucleosome positioning. The RSC complex facilitates the transfer of a histone octamer from a nucleosome core particle to naked DNA, a process requiring ATP and involving an activated RSC-nucleosome intermediate. This remodeling reaction also includes DNA translocation, DNA twisting, and conformational changes. As a reconfigurer of centromeric and flanking nucleosomes, the RSC complex is essential for proper kinetochore function in chromosome segregation. Additionally, via a PKC1-dependent signaling pathway, it contributes to the organization of the cellular cytoskeleton. This subunit is required for the transcription of ribosomal protein genes and genes involved in maintaining the integrity of the cell wall. In conjunction with HTL1, LDB7, NPL6, and RSC3 components, RSC30 defines a fungal-specific module within the RSC complex, playing a role in various cellular functions, including the maintenance of cell wall integrity.
