SUMO3L is a ubiquitin-like protein that can be covalently attached to target lysine residues, either as a monomer or as a lysine-linked polymer. Unlike ubiquitin, SUMO3L does not appear to be involved in protein degradation and may act as an antagonist of ubiquitin in this process. SUMO3L plays a role in a variety of cellular functions, including nuclear transport, DNA replication and repair, mitosis, and signal transduction. Attachment to its substrates requires prior activation by the E1 complex SAE1-SAE2 and subsequent linkage to the E2 enzyme UBE2I.
