Description

Target Protein: SMTNL2 (Smoothelin-Like 2)

SMTNL2 is a 50 kDa protein encoded by the SMTNL2 gene (NCBI Gene ID: 342527). It is expressed in smooth muscle tissues and plays roles in:

  • Modulating myosin phosphatase activity .

  • Regulating calcium sensitivity in smooth muscle contraction .

  • Involvement in metabolic pathways, including glucose uptake and insulin signaling .

3.1. Tissue and Cell Line Reactivity

The antibody detects SMTNL2 in:

  • Mouse small intestine tissue .

  • Human cell lines: MCF-7 (breast cancer), COLO 320 (colorectal cancer), and SGC-7901 (gastric cancer) .

3.2. Key Findings in Research

  • Smooth Muscle Function: SMTNL2 modulates myosin light-chain phosphatase, influencing smooth muscle tone .

  • Cancer Relevance: Elevated SMTNL2 expression correlates with tumor progression in colorectal and gastric cancers .

  • Subcellular Localization: Predominantly cytoplasmic, with dynamic redistribution during cellular stress .

4.1. Western Blot

  • Detects a ~68–70 kDa band in human, mouse, and rat lysates (Fig. 1A) .

  • Positive controls: COLO 320 (colorectal cancer) and MCF-7 cells .

4.2. Immunofluorescence

  • Localizes SMTNL2 to the cytoplasm in MCF-7 cells (Fig. 1B) .

4.3. Immunoprecipitation

  • Efficiently pulls down SMTNL2 from COLO 320 cell lysates .

Limitations and Considerations

  • Post-Translational Modifications: Observed molecular weight discrepancies (68–70 kDa vs. predicted 50 kDa) require careful interpretation .

  • Cross-Reactivity: Untested in non-mammalian systems .

Target Background

Function
SMT2 catalyzes the transfer of a methyl group from S-adenosyl-methionine to the methylene group of 24-methylene lophenol, resulting in the formation of 24-ethylidene lophenol.
Gene References Into Functions
  1. SMT3 exhibits functional redundancy with SMT2. Organisms lacking both SMT1 and SMT2 are embryonically lethal. A simultaneous mutation in SMT2 and SMT3 leads to significant alterations in sterol composition. PMID: 20421456
