Sorting Nexin 5 (SNX5) is a protein that plays a crucial role in various stages of intracellular trafficking. It interacts with membranes containing phosphatidylinositol 3-phosphate (PtdIns(3P)) or phosphatidylinositol 3,4-bisphosphate (PtdIns(3,4)P2). SNX5 is a component of the retromer membrane-deforming SNX-BAR subcomplex, which mediates retrograde transport of cargo proteins from endosomes to the trans-Golgi network (TGN). This process is essential for endosome-to-plasma membrane transport and cargo protein recycling. The SNX-BAR subcomplex deforms the donor membrane into a tubular profile known as the endosome-to-TGN transport carrier (ETC). SNX5 is involved in retrograde transport of lysosomal enzyme receptor IGF2R and may act as a link between endosomal transport vesicles and dynactin. It also contributes to the internalization of epidermal growth factor receptor (EGFR) after EGF stimulation. SNX5 plays a role in EGFR endosomal sorting and degradation, a process involving PIP5K1C isoform 3 and independent of the retromer pathway. SNX5, in conjunction with PIP5K1C isoform 3, facilitates the interaction of HGS with ubiquitinated EGFR, leading to EGFR sorting into the intraluminal vesicles (ILVs) of the multivesicular body for subsequent lysosomal degradation.
