IleS is an enzyme that catalyzes the attachment of isoleucine to tRNA(Ile). Due to its ability to inadvertently accommodate and process structurally similar amino acids like valine, IleS possesses two distinct tRNA(Ile)-dependent editing activities to prevent such errors. One activity, known as 'pretransfer' editing, involves the hydrolysis of activated Val-AMP. The other activity, designated as 'posttransfer' editing, involves deacylation of mischarged Val-tRNA(Ile).
