Function
The EcoprrI methyltransferase (MTase) complex comprises M and S subunits, which together methylate two adenine residues within a bipartite DNA recognition sequence on complementary strands. In the presence of the R subunit, the complex exhibits endonuclease activity, binding to the same target sequence but cleaving the DNA at a distance from this site. The enzyme's function (methylation or cleavage) is determined by the methylation status of the target sequence. Unmodified target sites result in DNA cleavage, while hemimethylated sites are modified by the complex's maintenance MTase activity, leading to methylation of both strands. The S subunit dictates the enzyme's DNA sequence specificity; EcoprrI recognizes the sequence 5'-CCAN7ATGC-3'.
