Target Background

Function
UBP16 is a deubiquitinating enzyme that specifically recognizes and hydrolyzes the peptide bond at the C-terminal Gly of ubiquitin. It plays a crucial role in the processing of poly-ubiquitin precursors and ubiquitinated proteins. Moreover, UBP16 is involved in salt tolerance by regulating sodium transport activity and suppressing cell death, at least partially, by modulating the stability and activity of SHM1. Additionally, UBP16 contributes to cadmium tolerance through its interaction with HIPP27, potentially affecting the stability of HIPP27.

FAQs for Researchers on USP16 Antibody Applications in Academic Research

Advanced Research Questions

  • How can USP16 antibody experimental design address contradictory findings in DNA repair studies?
    Discrepancies often arise from:

    • Cellular context: USP16 exhibits substrate specificity dependent on cell type (e.g., ovarian vs. pancreatic cancer) .

    • Post-translational modifications: USP16 activity is regulated by phosphorylation during mitosis .
      Mitigation strategy:

    • Include isoform-specific antibodies (e.g., targeting phosphorylated vs. unmodified USP16).

    • Combine in vitro (recombinant protein) and in vivo (tissue microarray) validation .

  • What are optimal protocols for studying USP16’s role in immune dysregulation?
USP16 modulates NF-κB signaling by deubiquitinating IKKβ, impacting cytokine production . Key steps:

    • Model systems: Use USP16-deficient murine models or patient-derived immune cells.

    • Functional assays: Measure cytokine levels (e.g., IFN-α) post-USP16 inhibition .

    • Multi-omics integration: Correlate USP16 activity with transcriptomic/proteomic profiles .
