Description

Localization Studies

  • M-lines: UNC-98 colocalizes with UNC-89 (obscurin homolog) at sarcomeric M-lines, critical for thick filament stability .

  • Nuclei: Detected in muscle nuclei under specific fixation conditions (e.g., n-heptane) .

  • Dense Bodies: Observed in rescued unc-98 mutants overexpressing UNC-98 .

Functional Insights

  • Muscle Phenotypes: unc-98 mutants exhibit disorganized A-bands, shortened M-lines, and paramyosin-rich cytoplasmic aggregates .

  • Genetic Interactions:

    • UNC-97 (PINCH homolog): Direct interaction via yeast two-hybrid assays; UNC-98 stabilizes UNC-97 localization .

    • Paramyosin (UNC-15): UNC-98 promotes paramyosin incorporation into thick filaments and prevents aggregation .

Target Background

Function
UNC-98 is a probable transcription factor essential for muscle structure. Its dual subcellular localization (both cytoplasmic and nuclear) suggests a bifunctional role: it may function as a muscle adhesion complex protein and/or as a transcription factor (or in concert with transcription factors) to modulate gene expression. It is hypothesized to act as a molecular bridge between UNC-97 and MYO-3 at the muscle M-line, potentially mediating signaling pathways. Furthermore, UNC-98 plays a crucial role in the formation of muscle connections—specifically, muscle arm extensions—between the body wall and motor axons within the dorsal and ventral nerve cords.
Gene References Into Functions
UNC-98 functions as both a structural component of muscle focal adhesions and a nuclear protein influencing gene expression. [PMID: 12808046](https://www.ncbi.nlm.nih.gov/pubmed/12808046)

Subcellular Location
Nucleus. Cytoplasm. Note=Localized in body wall muscle M-lines, dense bodies and muscle nuclei.
Tissue Specificity
Expressed in embryos from 1.5- to 2-fold stage in myofibrils. In larvae and adults, it is expressed in body wall muscle, and in addition, anal depressor muscle and vulval muscles. More specifically it is found in the thick filaments of muscle fibers.
