Description

Biological Roles of VAMP3

VAMP3 facilitates membrane fusion in diverse cellular processes:

  • Endosomal Recycling: Mediates recycling of transferrin receptors to the plasma membrane .

  • Platelet Function: Required for α-granule secretion during platelet activation .

  • Immune Response: Regulates T-cell receptor recycling to immunological synapses .

  • Cell Migration: Coordinates membrane trafficking during motility .

Mechanistic Insights

  • VAMP3 knockout studies revealed impaired granule secretion in platelets, linking it to hemostasis and thrombosis .

  • In T-cells, VAMP3 depletion disrupted receptor recycling, impairing immune synapse formation .

Disease Associations

  • Cancer: Elevated VAMP3 expression correlates with metastatic potential in breast cancer models .

  • Neurological Disorders: Altered VAMP3 levels are observed in synaptic dysfunction pathologies .

Validation and Technical Considerations

  • Western Blot: Detects endogenous VAMP3 at ~13 kDa in human, mouse, and rat lysates .

  • Immunoprecipitation: Efficiently isolates VAMP3 complexes for interactome studies .

  • Storage: Stable at -20°C for 12 months; avoid freeze-thaw cycles .

Target Background

Function
VAM3 Antibody is essential for vacuolar assembly. It provides the t-SNARE function in a late stage of this process. This antibody is also required for homotypic vacuole membrane fusion, autophagy, and fusion of biosynthetic transport vesicles with the vacuole. Furthermore, it plays a crucial role in the delivery of alpha-factor receptor-ligand complexes to the vacuole.
