Target Background

Function
Vanin-1 (VNN1) is an amidohydrolase enzyme that specifically hydrolyzes one of the carboamide linkages in D-pantetheine. This activity effectively recycles pantothenic acid (vitamin B5) and releases cysteamine.
Gene References Into Functions
  • Research suggests that the metabolic pathway catalyzed by VNN1 pantetheinase plays a suppressive role in influenza A virus (IAV) infection of the respiratory tract, particularly in severe cases characterized by hypercytokinemia. PMID: 28576495
  • Data indicates that the G protein-coupled receptor, family C, group 5, member A protein (GPRC5A) regulates oxidative stress through the vanin 1 protein (VNN1). PMID: 28316092
  • Studies suggest that urinary vanin-1 serves as an early predictive biomarker for a decline in estimated glomerular filtration rate (eGFR) in patients with urothelial carcinoma after cisplatin treatment. PMID: 27317936
  • Serum pantetheinases, encoded by the VNN1 gene, regulate erythrocyte lifespan and influence the risk of developing complicated malaria. PMID: 26343328
  • VNN1 contributes to corticosteroid responsiveness. Variations in VNN1 nasal epithelial mRNA expression and VNN1 promoter methylation could serve as clinically useful biomarkers for treatment response in asthmatic children. PMID: 25910714
  • A study provides compelling biological evidence for the association between a specific SNP and blood pressure. The study suggests that vanin-1 misfolding and degradation are the underlying molecular mechanisms. PMID: 25233454
  • Research indicates that hepatic vanin-1 is highly sensitive to regulation by peroxisome proliferator-activated receptor alpha (PPARalpha). Plasma vanin activity could potentially serve as a readout of changes in PPARalpha activity in human subjects. PMID: 24751833
  • The X-ray crystal structure of human vanin 1 at a resolution of 2.25 angstroms has been determined. This is the first reported structure from the vanin family, and it includes a crystal structure of vanin 1 bound to a specific inhibitor. PMID: 25478849
  • Serum vanin-1 levels may be low in end-stage renal failure and transiently increase after transplantation due to transient renal function deterioration, which does not lead to an elevation of serum creatinine levels in renal transplant patients. PMID: 24702142
  • Research suggests that vanin-1 can distinguish between chronic responders and non-responders in patients with immune thrombocytopenia (ITP) as well as between newly diagnosed ITP patients and healthy controls. PMID: 23534352
  • Alternative splicing in the VNN1 gene has been associated with colorectal cancer. PMID: 24687856
  • Single-nucleotide polymorphisms (SNPs) in the regulatory regions of the VNN1 gene have been linked to inflammatory bowel diseases. PMID: 23949622
  • Microparticle internalization requires the interaction of the ectoenzyme Vanin-1 (VNN1), a prevalent surface protein on microparticles, with lipid raft domains of endothelial cells. PMID: 24106341
  • Vanin-1 is the enzyme that recycles pantothenic acid (vitamin B5) generated during coenzyme A breakdown and is actively involved in the progression of inflammatory reactions by generating cysteamine. PMID: 23978960
  • The liver contributes to vanin-1 secretion in serum, and PPARalpha is a limiting factor in serum Vnn1 production. PMID: 24140347
  • Data do not support a significant role for the Vanin 1 T26I variant in determining blood pressure levels and incident ischemic events. PMID: 21550219

  • Urinary vanin-1, an ectoenzyme pantetheinase, effectively differentiated diabetic patients with macroalbuminuria from those with normal albuminuria. PMID: 21544065
  • Vanin-1 is a specific and sensitive biomarker for renal tubular injury induced by organic solvents. PMID: 21907259
  • Overexpression of VNN1, an oxidative stress sensor in epithelial cells, was strongly associated with progression to chronic immune thrombocytopenia. PMID: 21325602

  • The missense SNP rs2272996 (or N131S) in the VNN1 gene was significantly associated with hypertension in African Americans and this association was replicated in Mexican Americans. A non-significant opposite association was observed in European Americans. PMID: 18043751
  • Vanin-1 and vanin-3 are induced at the mRNA and protein levels by psoriasis-associated proinflammatory cytokines (Th17/Th1) but not by Th2 cytokines. PMID: 19322213
