Product Specs

Introduction
Interleukin-27 (IL-27) is a heterodimeric cytokine composed of two subunits, IL-27p28 and EBI3 (EBV induced gene 3). It plays a crucial role in regulating immune responses, particularly in the context of T cell activation and differentiation. IL-27 binds to its receptor complex, WSX1/TCRR, and initiates downstream signaling pathways. It exhibits both pro-inflammatory and anti-inflammatory properties depending on the cellular context and disease model.
Description
Recombinant Mouse Interleukin-27 is produced in E. coli. This non-glycosylated polypeptide chain consists of 207 amino acids with a molecular weight of 23.7 kDa. The purification process involves proprietary chromatographic techniques.
Physical Appearance
Sterile Filtered White lyophilized (freeze-dried) powder.
Formulation

Lyophilized from a 10mM NaHCO₃ buffer with a pH of 8.5.

Solubility

Reconstitute the lyophilized IL-27 in sterile 18MΩ-cm H2O to a concentration of at least 100µg/ml. Further dilutions can be made in other aqueous solutions.

Stability
Lyophilized IL-27 is stable at room temperature for 3 weeks. However, it is recommended to store the product desiccated below -18°C. After reconstitution, store IL-27 at 4°C for 2-7 days. For long-term storage, keep it below -18°C. Minimize freeze-thaw cycles.
Purity

Purity is determined to be greater than 95.0% by SDS-PAGE analysis.

Biological Activity

The biological activity of Mouse p28, a subunit of IL-27, is assessed by measuring its ability to inhibit IL-17A expression in mouse CD4 splenocytes stimulated with TGF-beta and IL-6. A concentration of 50ng/ml of mouse p28 can inhibit IL-17A expression by over 25% in this assay.

Q&A

FAQs on IL-27 in Mouse Models for Academic Researchers

Advanced Research Questions

How does IL-27 exhibit contradictory roles in autoimmune vs. infectious contexts?

ContextPro-inflammatory RoleAnti-inflammatory Role
ColitisPromotes T cell-driven inflammation Suppresses Th17 responses in EAE
Neuropathic PainN/AInhibits pain via IL-10 induction
InfectionLimits S. aureus clearance (via IL-10) Enhances survival in viral encephalitis

What mechanisms explain IL-27’s dual regulation of T cells?

  • Pro-inflammatory:

    • Suppresses Foxp3+ Treg differentiation, exacerbating colitis .

    • Enhances IFN-γ production in Th1 cells during Mycobacterium tuberculosis infection .

  • Anti-inflammatory:

    • Induces IL-10 in Tr1 cells to mitigate EAE .

    • Inhibits Th17 differentiation via RORγt suppression .

How do species-specific differences in IL-27 biology affect translational research?

  • Structural divergence:

    • Murine IL-27α (p28) is secreted alone and functions independently, unlike human IL-27α, which requires EBI3 for stability .

  • Functional implications:

    • Mouse models may overestimate IL-27α’s standalone role in immune regulation compared to humans.

Methodological Challenges

How to address contradictory findings in IL-27’s role in neuroinflammation?

  • Experimental design tips:

    • Use conditional knockout mice (e.g., T cell-specific Il27ra deletion) to isolate cell-type-specific effects .

    • Combine cytokine profiling (e.g., Luminex) with single-cell RNA sequencing to map IL-27-responsive subsets .

  • Data interpretation: Context-dependent effects (e.g., acute vs. chronic inflammation) require longitudinal studies .

What are pitfalls in interpreting IL-27 knockout phenotypes?

  • Compensatory mechanisms:

    • EBI3⁻/⁻ mice show upregulated IL-35 (another EBI3-containing cytokine), confounding results .

  • Solution: Pair knockout models with neutralizing antibodies (e.g., anti-IL-35) .

Product Science Overview

Structure and Composition

IL-27 consists of two subunits:

  1. EBI3 (Epstein-Barr virus-induced gene 3): This subunit is related to the IL-12p40 protein.
  2. p28 (also known as IL-30): This subunit is related to the IL-12p35 polypeptide .

These subunits form a complex that is essential for the cytokine’s biological activity. The recombinant mouse IL-27 is produced in E. coli and is a single, non-glycosylated polypeptide chain containing 207 amino acids, with a molecular mass of approximately 23.7 kDa .

Biological Activity

IL-27 has several important functions in the immune system:

  • Activation of Naïve T Cells: IL-27 signaling occurs via a receptor complex composed of WSX-1 and glycoprotein (gp) 130. This signaling leads to the activation and phosphorylation of STAT1 and STAT3 in naïve CD4+ T cells .
  • Proliferation of Erythroleukemic Cells: Recombinant mouse IL-27 induces the proliferation of TF-1 human erythroleukemic cells in a dose-dependent manner .
  • Inhibition of IL-17A Expression: Mouse p28, a component of IL-27, inhibits TGF-beta and IL-6-induced IL-17A expression in mouse CD4 splenocytes .
Applications and Usage

Recombinant mouse IL-27 is used in various research applications, including:

  • Immunological Studies: To understand the role of IL-27 in immune regulation and its potential therapeutic applications.
  • Cell Proliferation Assays: To study the effects of IL-27 on cell proliferation and signaling pathways.
Storage and Handling

For optimal results, recombinant mouse IL-27 should be stored at -20°C or colder. It is stable at room temperature for up to three weeks when lyophilized but should be stored desiccated below -18°C for long-term storage. Upon reconstitution, it should be stored at 4°C for short-term use .
