Introduction
Dendritic cell and monocyte chemokine-like protein (DMC/CXCL17/VEGF-correlated chemokine 1/VCC1) is a secreted molecule structurally similar to chemokines CXCL8/IL8 and CXCL14/BRAK. Primarily expressed by airway and intestinal epithelium, CXCL17 selectively attracts and binds quiescent, but not activated, peripheral blood monocytes and dendritic cells. Elevated CXCL17 expression is observed during in vitro tube formation in endothelial cells. Moreover, CXCL17, along with CXCL1/GRO and CXCL8/IL8, demonstrates a significant expression correlation with VEGF in primary lung, breast, and esophageal tumors, suggesting its involvement in tumor angiogenesis. Rat CXCL17 exhibits 82% and 71% amino acid sequence identity with its mouse and human counterparts, respectively.
Description
Recombinant Rat CXCL17, expressed in E. coli, is a non-glycosylated polypeptide chain with a molecular weight of 11.5 kDa, consisting of 97 amino acids. The protein is purified using proprietary chromatographic techniques.

Sterile white lyophilized powder.
Formulation
The CXCL17 protein was lyophilized from a 0.2 μm filtered solution containing 20 mM phosphate buffer (PB), 300 mM sodium chloride (NaCl), at pH 7.4.
Solubility
Reconstitute the lyophilized CXCL17 in sterile 18 MΩ-cm water to a concentration of at least 100 µg/ml. This solution can be further diluted in other aqueous solutions.
Stability
Lyophilized CXCL17 remains stable at room temperature for up to 3 weeks. However, for long-term storage, it is recommended to store the lyophilized protein desiccated at temperatures below -18°C. After reconstitution, the CXCL17 solution should be stored at 4°C for no longer than 2-7 days. For future use, store the solution at -18°C. The addition of a carrier protein (0.1% HSA or BSA) is advised for extended storage. Avoid repeated freeze-thaw cycles.
Purity
The purity of CXCL17 is greater than 95%, as determined by: (a) Reverse-phase high-performance liquid chromatography (RP-HPLC) analysis and (b) Sodium dodecyl-sulfate polyacrylamide gel electrophoresis (SDS-PAGE) analysis.
Biological Activity
The biological activity of CXCL17 is evaluated by its ability to stimulate VEGF expression in mouse endothelial cells. The effective concentration for inducing this effect (ED50) typically falls within the range of 1-5 µg/ml.

VCC1 is a secreted molecule with a size and predicted 3-dimensional folding pattern similar to that of chemokines CXCL8/IL8 and CXCL14/BRAK . It is constitutively expressed in the lung and can also be detected in the trachea, stomach, and skeletal muscle . The mature Rat CXCL17 shares 82% and 71% amino acid sequence identity with mouse and human CXCL17, respectively .

Function and Biological Activity

VCC1 plays a significant role in the chemotaxis of quiescent peripheral blood monocytes and dendritic cells, but not LPS-activated ones . It specifically binds to these cells and induces their migration. The expression of VCC1 is increased in endothelial cells when they are induced to form tubes in vitro . This chemokine is also involved in angiogenesis, showing significantly correlated expression with VEGF in primary lung, breast, and esophageal tumors . Therefore, VCC1 is suggested to have a role in tumor angiogenesis.

Recombinant Production

Recombinant VCC1 is produced in Escherichia coli (E. coli) as a single, non-glycosylated polypeptide chain containing 98 amino acids and having a molecular mass of 11.5 kDa . Stability and Storage

Lyophilized VCC1 is stable at room temperature for three weeks but should be stored desiccated below -18°C for long-term storage .
